ABSTRACT
INTRODUCTION: Oxidative stress plays a critical role in ovarian failure and menopause-related disorders. Malondialdehyde (MDA) and glutathione (GSH) are key markers of oxidative damage and antioxidant defense.
OBJECTIVE(S): This study aimed to assess the antioxidant potential of SKF7 in VCD- induced ovarian failure by measuring MDA and GSH levels in ovarian tissue.
MATERIALS & METHODS: Female rats were divided into seven groups: Sham + H2O, Sham + 500 mg/kg BW SKF7, VCD + H2O, VCD + SKF7 (125, 250, and 500 mg/kg BW), and VCD + estradiol. VCD was administered to induce ovarian failure. Treatments were given orally for 65 days. Ovarian tissues were analyzed for MDA and GSH concentrations using spectrophotometric assays. VCD exposure significantly increased MDA levels and reduced GSH concentrations, indicating oxidative stress.
RESULTS: SKF7 treatment, particularly at 250 mg/kg, significantly decreased MDA levels and restored GSH levels compared to the VCD + H20 group. The 250 mg/kg SKF7 group showed comparable antioxidant activity to the estradiol-treated group. Sham groups maintained baseline oxidative profiles. SKF7 demonstrated dose- dependent antioxidant effects, mitigating VCD-induced oxidative stress in ovarian tissue. The 250 mg/kg dose was the most effective in restoring redox balance without exceeding normal baseline levels observed in sham groups.
CONCLUSION: SKF7 exhibits promising antioxidant properties in the rat model suggesting potential as a supportive herbal therapy for oxidative stress-related reproductive disorders.
